    miRNA Expression in Colon Polyps Provides Evidence for a Multihit Model of Colon Cancer

    Changes in miRNA expression are a common feature in colon cancer. Those changes occurring in the transition from normal to adenoma and from adenoma to carcinoma, however, have not been well defined. Additionally, miRNA changes among tumor subgroups of colon cancer have also not been adequately evaluated. In this study, we examined the global miRNA expression in 315 samples that included 52 normal colonic mucosa, 41 tubulovillous adenomas, 158 adenocarcinomas with proficient DNA mismatch repair (pMMR) selected for stage and age of onset, and 64 adenocarcinomas with defective DNA mismatch repair (dMMR) selected for sporadic (n = 53) and inherited colon cancer (n = 11). Sporadic dMMR tumors all had MLH1 inactivation due to promoter hypermethylation. Unsupervised PCA and cluster analysis demonstrated that normal colon tissue, adenomas, pMMR carcinomas and dMMR carcinomas were all clearly discernable. The majority of miRNAs that were differentially expressed between normal and polyp were also differentially expressed with a similar magnitude in the comparison of normal to both the pMMR and dMMR tumor groups, suggesting a stepwise progression for transformation from normal colon to carcinoma. Among the miRNAs demonstrating the largest fold up- or down-regulated changes (≥4), four novel (miR-31, miR-1, miR-9 and miR-99a) and two previously reported (miR-137 and miR-135b) miRNAs were identified in the normal/adenoma comparison. All but one of these (miR-99a) demonstrated similar expression differences in the two normal/carcinoma comparisons, suggesting that these early tumor changes are important in both the pMMR- and dMMR-derived cancers. The comparison between pMMR and dMMR tumors identified four miRNAs (miR-31, miR-552, miR-592 and miR-224) with statistically significant expression differences (≥2-fold change)

    Exploring disturbance as a force for good in motor learning

    Disturbance forces facilitate motor learning, but theoretical explanations for this counterintuitive phenomenon are lacking. Smooth arm movements require predictions (inference) about the force-field associated with a workspace. The Free Energy Principle (FEP) suggests that such 'active inference' is driven by 'surprise'. We used these insights to create a formal model that explains why disturbance might help learning. In two experiments, participants undertook a continuous tracking task where they learned how to move their arm in different directions through a novel 3D force field. We compared baseline performance before and after exposure to the novel field to quantify learning. In Experiment 1, the exposure phases (but not the baseline measures) were delivered under three different conditions: (i) robot haptic assistance; (ii) no guidance; (iii) robot haptic disturbance. The disturbance group showed the best learning as our model predicted. Experiment 2 further tested our FEP inspired model. Assistive and/or disturbance forces were applied as a function of performance (low surprise), and compared to a random error manipulation (high surprise). The random group showed the most improvement as predicted by the model. Thus, motor learning can be conceptualised as a process of entropy reduction. Short term motor strategies (e.g. global impedance) can mitigate unexpected perturbations, but continuous movements require active inference about external force-fields in order to create accurate internal models of the external world (motor learning). Our findings reconcile research on the relationship between noise, variability, and motor learning, and show that information is the currency of motor learning

    Storm-time response of the mid-latitude thermosphere: Observations from a network of Fabry-Perot interferometers

    Observations of thermospheric neutral winds and temperatures obtained from a network of five Fabry-Perot interferometers deployed in the midwest United States during a geomagnetic storm on 2 October 2013 showed that coincident with the commencement of the storm, the horizontal wind was observed to surge westward and southward (towards the equator). Simultaneous with this surge in the horizontal winds, an apparent downward wind of approximately 100 m/s lasting for 6 hours was also observed. The neutral temperature was observed to increase by approximately 400 K over all of the sites. Similar results of downward vertical winds and sustained heating have been seen in other geomagnetic storm events. The large sustained apparent downward winds are interpreted as arising from the contamination of the nominal spectral profile of the 630.0-nm population distribution, which is thermalized within the thermosphere region, by fast O related to the infusion of low-energy O+ ions that are generated by charge exchange and momentum transfer collisions. This interpretation is supported through simultaneous observations made by the Helium, Oxygen, Proton, and Electron spectrometer instruments on the twin Van Allen Probes spacecrafts, which show an influx of low-energy ions well correlated with the period of apparent downward winds. These results emphasize the importance of distributed networks of instruments in understanding the complex dynamics that occur in the upper atmosphere during disturbed conditions and represent an example of magnetosphere-ionosphere coupling
